NEW MATERIAL:A human monocyto growth factor capable of inducing differentiation and growth of human monocyto. USE:An infection preventive, an immunostimulant and anticancer agent. PREPARATION:For example, a culture filtrate of human ling cancer cell strain T3M-30Lu is collected and, after removal of low molecular weight components by a hollow fiber filter, concentrated and then adjusted to pH7.5. Ammonium sulfate is added thereto and the precipitate generated by the saturated ammonium sulfate of 30-60 wt.% is collected. The collected precipitate is dissolved in a trishydrochloric acid buffer solution (pH7.5) and substituted to the gel filtration method for collection of fractions having a monocyto growth factor activity. The collected fractions are subjected to the ion-exchange chromatography and then eluted by a NaCl concentration slope of 0-200mM and the resultant active fractions are subjected to the reversed phase high performance liquid chromatography and the eluted by a liner concentration slope of 0-99wt.% acetonitrile against 0.1wt.% trifluoroacetic acid, thus obtaining the objective human monocyto growth factor consisting of a proteinaceous substance having 21000 molecular weight and capable of inducing differentiation and growth of human monocyto. |
