NEW MATERIAL:The title antibody having the following characteristics:(1) capable of specifically binding to pyocyanic exotoxin A (toxin A), (2) capable of neutralizing the cytotoxicity induced by at least one molecule of the toxin A in a cell culture system, (3) when parenterally administered in vivo in a mouse at a level of 4X10<-12>mol, being capable of neutralizing in vivo the lethal toxicity induced by 15X10<-12>mol of the toxin A, resulting in a survival of >=50%, (4) capable of recognizing as an epitope a segment of the formula corresponding to the 591th to 613th amino acid residues of the toxin A, (5) classified as IgG3, and (6) giving optimal binding activity to the toxin A at pH >=5 with the resultant bond stable at pH 3. USE:Effecitve for the therapies and prevention of pyocyanic infectious diseases with the toxin A involved therein, having synergistic effects with antibiotics. PREPARATION:Human B lymphocytes spontaneously sensitized with the toxin A in a pyocyanic infectious disease is infected with Epstein-Barr virus, and the resulting transformant cells are fused with myeloma cells. Thence, the resultant hybridoma is incubated to produce the objective antibody. |
