PURPOSE:A plasmid pCG 2 capable of becoming a clone vector for a bacterium belonging to the genus Corynebacterium or its relative mold useful for preparing amino acid, nucleic acid, etc. obtained from a culture mold prepared by cultivating Corynebacterium glutamicum 225-218. CONSTITUTION:A plamid pCG 2 having a molecular weight of about 6.6 kilo base, sensitivity sites against restricted enzyme of Hind III 3, KpnI2, BamHI1, PstI1, HpaI1, EcoR 0, SalI0, BglII 0. Corynebacterium glutamicum 225-218 strain (FERM-P 5954) is cultivated in an NB medium, a mold is collectd from the culture solution, it is processed to have sensitivity to the white lysozyme, and dissolved in a cell wall by lysozyme, and a plasmid pCG 2 is separated from the bacteriolysin. |
