A glucoamylase, which had max. activity at ~65≦̸C, was isolated from a stachybotrys subsimplex culture. Thus, S. subsimplex G 30-1140 was aerobically cultured at 30≦̸C for 7 days on a medium contg. sol. starch 5, corn ext. 2, cotton oil sludge 0.5, yeast ext. 0.5, K2HPO4 0.1, MgSO4 0.05, and CaCl2 0.01%. Glucoamylase was isolated from the culture broth. The glucoamylase was purified by DEAE-cellulose and 0.05M Tris-HCl buffer contg. 1mM EDTA. When the resulting glucoamylase was treated with 2% maltodextrin soln. at pH 6.0 for 10 min., max, activity was observed at ~65≦̸C.
