PURPOSE:To enable to enhance the productivity of the subject substance and secrete the substance outside the cells by culturing a specific microorganism in a medium, the specific microorganism holding a recombinant DNA containing a gene coding a specified cholesterol.oxydase. CONSTITUTION:A DNA fragment containing a cholesterol.oxydase gene is cloned from the chromosome of a microorganism belonging to the genus Brevibacterium, and the produced chromosome DNA is recombined with a vector DNA (a plasmid capable of living in the Escherichia coli as a host) to produce a recombinant DNA. The recombinant DNA is introduced into a bacterium belonging to the Streptomyces to produce Streptomyces.lividans CHS31 strain (FERN BP-2969). The produced microorganism is cultured in a nutritive medium, etc., and the cells are subsequently removed from the culture solution. The obtained culture supernatant is treated by a conventional method to provide pure cholesterol.oxydase, thereby permitting to provide the objective substance in an extremely high purity and at a low cost by the fermentation process. |
