PURPOSE:To provide a novel esterase hydrolyzing the ester bond of an organic carboxylic acid ester, activated with Ca ions and having specific physical properties by culturing Serratia marcescens belonging to the genus Serratia microorganisms. CONSTITUTION:Serratia marcescens Sr41 (FERN No.41) belonging to the genus Serratia microorganisms is cultured, and the supernatant of the culture solution is salted out with a 45% saturated ammonium sulfate. The produced precipitates are separated, dissolved in a tris buffer solution (pH 7.5) and purified with an anion exchange resin by a gel filtration method, etc., to provide a novel esterase which acts on organic carboxylic acid alkyl esters, triglycerides and thiol esters, having an optimal pH of 7.5-9.0 and an optimal temperature of 40-50 deg.C when an olive oil is used as a substrate, stable at a pH of 8.0 below 50 deg.C, having a mol.wt. of 62000+ or -2000 (SDS-PAGE method) and an isoelectric point of 4.6+ or -0.1, activated with 1mM of Ca ions and inhibited with 1mM of Co, Ni or Fe ions and with EDTA. |
