Detection of sulphated oligo- and polysaccharide(s) - using poly base, and methylene blue opt. with an oxidising agent or auramine

An electrophoretic process for the detection of sulphated oligo and polysaccharides and their derivs. uses, for the detection reaction, a poly base, methylene blue opt. in combination with an oxidising agent or auramine. The polybase is lysine octadecyl ester, polylysine or polydimethyl amino ethyl aspartamide, and is esp. labelled with a fluorescence label or a radio label. Methylene blue is used opt. with ammonium persulphate as the oxidising agent. A surfactant is used in the electrophoresis. A gel with molecular sieve effect is added, esp. a gel with a polyacrylamide conc. of at least 10%. USE/ADVANTAGE - The process is rapid and simple and can be used clinically. Unlabelled sulphated saccharides can be quantitatively and qualitatively determined, and any structural changes in these cpds. can also be detected. The process can therefore be used in the monitoring of treatment of AIDS using sulphated oligosaccharides and to determine the correct dose needed and suitable intervals between doses.
