A method for preparing an extract of ginseng is provided to increase the content of ginsenoside Rg3 and Delta20-ginsenoside Rg3 known to have excellent anti-tumor and anti-cancer effect compared with other ginsenoside derivatives and utilize lactic acid bacteria and/or fermentation enzyme existing in enterobacteria by ripening a culture solution at a specific temperature for a certain time, and obtain the ginseng extract without heat-treatment, neutralization and extraction purification. A method for preparing an extract of ginseng comprises the steps of: (a) fermenting ginseng with lactic acid bacteria or enterobacteria; and (b) ripening a culture solution obtained from the step(a) at a temperature of 50-85 deg.C for 4-12 hours. The method further comprises a step of concentrating or freeze-drying a ripened solution obtained from the step(b).
