The invention relates to methods for determining the inherited or acquired DNA repair capacity of a cell or cell population, and to the use of said method for determining the genetic susceptibility to cancer of a subject or for predicting the response of a subject suffering from cancer to a chemotherapy treatment directed to damaging the DNA.
These methods involve the use of a polynucleotide encoding in its natural form a fluorescent protein (e.g. EGFP). Said polynucleotide has been previously contacted with a genotoxic agent (e.g. UV), rendering said polynucleotide unable to express a functional fluorescent protein. Host repair system capacity will be assessed by fluorescence.
