<p>A complex of H - Ser - Tyr - Ser - Met-Glu(OH) - His - Phe - Arg - Try - Gly - Lys - Pro-Val - Gly - Lys - Lys - Arg - Arg - Pro - Val-Lys - Val - Tyr - ProOH (b 1-24 corticotropin) or is derivative by replacing -Glu(OH)- by -Glu(NH2)-, or an ester thereof, with a sparingly water-soluble Zn compound, in which complex at least 20 gm equivalents of Zn are present per gram mol of the tetracosapeptide, is prepared by treating the tetracosapeptide with an aqueous Zn salt solution containing at least 20 gm. equivalents of Zn per gram mol of peptide and with substantially the stoichiometric quantity, calculated on the Zn salt, of an alkali metal or ammonium compound capable of reacting with the Zn salt to produce a sparingly soluble Zn compound. Suitable Zn compounds are the hydroxide and phosphate. Pharmaceutical compositions comprise the above compounds in admixture with a carrier, or a mixture of the tetracosapeptide and a water-soluble Zn salt, at least 20 gm. equivalents of Zn being present per gram mol of the peptide, in admixture or conjunction with the stoichiometric quantity, calculated on the Zn salt, of an alkali metal or ammonium compound capable of reacting with the Zn salt to produce a sparingly water-soluble Zn compound. The compositions may be in the form of lyophilized ampoules containing sufficient NaCl to give an isotonic solution, or solutions, suspensions or emulsion for parenteral administration.</p> |
