1330900 Thiopeptin A4 FUJISAWA PHARMACEUTICAL CO Ltd 20 Nov 1970 [21 Nov 1969] 55430/70 Heading C2A The new antibiotic thiopeptin A4, which is a pale yellow crystalline solid melting with decomposition at 236‹ to 245‹ C.; containing C= 50À13%, H=5-30%, N=15À22%, S=12À02% and O by difference; having a molecular weight 1854; an optical rotation [α]23= - 81À5 degrees D (C=1À0 in CHCl 3 ); an Rf value of 0À57 on chromatography in ethyl acetate : n-hexane : 2N ammonia, 4 : 1 : 1 by volume; an ultraviolet absorption spectrum as shown in Fig. 1 having shoulders at 235 to 250 mÁ, 295 mÁ and 303 to 308 mÁ; and an infra-red spectrum as shovm in Fig. 2 having absorption peaks at 3340, 3330, 1735, 1655, 1585, 1530, 1490, 1420, 1345, 1305, 1250, 1210, 1160, 1110, 1090, 1065, 1025, 1000, 975, 950, 920, 895, 880, 860, 840, 768, 725, 705, 680 and 660 cm.<SP>-1</SP>; is produced by aerobically cultivating Streptomyces tateyamensis (ATCC. 21389) or a thiopeptin A4- producing variant or mutant thereof in an aqueous nutrient medium and recovering the thiopeptin A4 from the fermented medium. The antibiotic may be recovered by the same procedure as described in Specification 1,265,562 for the recovery of thiopeptin mixture (i.e. thiopeptins A 1 , A 2 , A 3 and B), such as extracting the fermentation broth with chloroform and chromatographing on a silicic acid column with chloroform : methanol, when the thiopeptin A4 is eluted as the last component. Animal feed compositions-Thiopeptin A4 may be employed as a growth-promoting additive in animal feedstuffs, especially chicken food. Thus, an animal feed or feed supplement may comprise a basal animal feed ration together with thiopeptin A4, which may be added in the form of a dried thiopeptin-A4-containing mycelium cake. |
