PURPOSE:To assay proteins unstable with respect to heat with a high recovery rate and a better reproducibility by measuring ultraviolet ray absorbance of a liquid eluted by tilt elution employing a shorter column filled with a silica gel treated by silane. CONSTITUTION:A shorter column filled with a silica gel treated by silane as column filler is combined with a mobile phase in which a strong acid aqueous solution of sodium dihydrogenphosphate is changed to a strong acid mixture of a sodium dihydrogenphosphate, ethyleneglycol and isopropanol to measure the ultraviolet ray absorbance of an eluted liquid by tilt elution at such a low temperature as about -20 deg.C-+20 deg.C. This enables the assay of proteins and peptides at a low temperature without denaturization. |
