PURPOSE:To provide a new microorganism transformed by a recombinant plasmid including a DNA coding for phospholipase A2, belonging to genus Streptomyces and having a function to stably produce phospholipase A2 in a large amount. CONSTITUTION:Streptomyces violaceoruber A-2688 (FER MP-13545) is cultured in a culture medium, the microbial cells are collected by the centrifugation of the cultured solution, the collected cells are dispersed in a buffered solution, lysozyme is added, cell walls are crushed, a surfactant is added, phenol/ chloroform (1/1) is added, the water phase is separated after mixing, ethanol is added to the water phase and DNA is obtained. A DNA library is established from the DNA. A clone containing the DNA of formula which codes for phospholipase A2 is selected from the DNA library. A recombinant plasmid is produced by combining the DNA into an expression vector. The recombinant plasmid is introduced into a microorganism belonging to genus Streptomyces and the objective new microorganism is obtained by the transformation. |
