A preparation method of endothelial dysfunction induced animal model is provided, which enables to develop various therapeutic agents by using the homocysteine-thiol lactone. A preparation method of endothelial dysfunction induced animal model comprises: a step(S110) for dissolving the homocysteine-thiol lactone in the phosphoric acid buffer and preparing it in solution; a step(S120) for cultivating the blood vessel endothelial cell; a step(S130) for inoculating a homocysteine-thiol lactone solution in the blood vessel endothelial cell culture fluid; and a step(S140) for confirming the apoptosis by using the MTT analysis after the homocysteine-thiol lactone addition of solution.
