<p>1,157,998. Antibiotic SL 1846. SANDOZ Ltd. 7 Nov., 1966 [6 Dec., 1965], No. 49846/66. Heading C2A. A cytostatic, antiviral and amebostatic antibiotic SL 1846, produced by the cultivation of Pseudeurotium ovalis NRRL 3194, is characterized by having the following properties: a colourless, crystalline neutral compound having the formula C 16 H 24 O 5 , a melting point of 89-92‹ C., a specific rotation of [α]<SP>20</SP> D = -117 degrees (C = 0À40 in chloroform), ultraviolet spectrum (Fig. 1, not shown) showing maximum at 284À5 mÁ (log # = 1À63) and strong adsorption at 210 mÁ (log E = 3À1) and infra-red spectrum as in Fig. 2. Antibiotic SL 1846 has the formula 4 - (2,3 epoxy - 6 - methyl - 5 - hepten - 2 - yl) - 4 - hydroxy - 5 - methoxy - 1 - oxaspiro -[2,5]- octan-6-one. The antibiotic is isolated from the clarified culture broth by extraction with a waterimmiscible solvent, e.g. ethylene chloride, benzene, ethyl acetate, butyl acetate, chloroform or butanol. Purification is by (a) chromatography on e.g. activated alumina, silica gel or magnesium silicate followed by elution with chloroform/methanol and (b) counter-current distribution. A pharmaceutical preparation for internal and external use comprises antibiotic SL 1846 and a carrier.</p> |
