Metoclopramide prepn - from p-amino- salicylic acid

Metoclopramide (I; N-(diethylaminoethyl)-4-amino-5-chloro-2-methoxybenzamide): a known cpd. useful in the treatment of functional disorders of the digestive tract, is prepd. in high yields by (a) reacting p-amino-salicylic acid with a methylating agent under anhydrous conditions, reacting the resulting methyl 2-methoxy-4-aminobenzoate (II) with H2NCH2CH2N(C2H5)2, and chlorinating the resulting N-(diethylaminoethyl)-2-methoxy-4-aminobenzamide with phenyl iododichloride.
