New compounds of the general formula <FORM:1078850/C2/1> where R is lower alkyl (1-8 C), lower alkenyl (2-8 C), phenyl-loweralkyl, methylphenylloweralkyl, cycloalkyl (3-9 C) or cycloalkenyl (3-9 C), R1 is phenyl optionally substituted by loweralkyl, diloweralkyl, alkoxy, halo, trifluoromethyl, loweralkyl and halo, loweralkylmercapto or diloweralkylamino, or is naphthyl, and R11 is halogen, hydroxy, mercapto, loweralkylmercapto, loweralkoxy, phenoxy, loweralkanoyloxy, cyano, carboxy, carbonyl halide, carblower alkoxy, or carbamyl or amino, including N-substitution derivatives, and certain acid addition and quaternary salts thereof, are prepared by reacting a compound of formula <FORM:1078850/C2/2> with carbonyl chloride or bromide to form an intermediate which is followed spontaneously or on heating by rearrangement to products wherein R11 is halogen, the other compounds being prepared therefrom, methods described being: reaction of halo compounds with appropriate alkali metal derivatives for alkoxy, cyano, alkanoyloxy and mercapto compounds, which last may be alkylated; basic hydrolysis of halo or carb-loweralkoxy for hydroxy; acid hydrolysis of nitriles for carboxy; halogenation of carboxy for carbonyl halide; esterification of the acid or its halide for carb-loweralkoxy; reaction of NH3 or amines with the acid alkyl ester or halide for carbamyl; reaction of NH3, amines or amides with halo for amino or substituted amino. Pyrrolidine intermediates II are prepared by reaction of R1NH2 with appropriate 3-halo- or 3 - alkyl or - aryl - sulphonyloxy compounds, said compounds being themselves obtained from the 1-R-3-hydroxy-pyrrolidines. Pharmaceutical compositions comprise the new compounds I with carriers, oral and parenteral preparations, useful as analeptics and hypotensives, being described. |
