Griseofulvin is produced by fermenting an aqueous nutrient medium, under aerobic conditions, with a griseofulvin-producing strain of Khuskia oryzae, Nigrospora oryzae, Nigrospora sphaerica, Nigrospora saccharii or Nigrospora musae, or a griseofulvin-producing mutant thereof, and isolating griseofulvin from the resulting fermentation broth. Suitable strains are Khuskia oryzae A.C.C. 3026; I.M.I. 122,490; Nigrospora oryzae A.C.C. 3145; and Nigrospora saccharii A.C.C. 3143. The fermentation may be carried out in still culture, shake culture, or submerged aerated deep culture. The medium should contain sources of assimilable carbon and nitrogen (e.g. carbohydrates or hydroxy acids, and nitrogen-containing inorganic salts, yeast extract or corn steep liquor solids) together with sources of phosphorus, magnesium, sulphur and potassium, and traces of Fe, Mn, Zn, Cu and Mo. Suitable conditions are pH 5À5-6À5 at 15-30‹ C. Griseofulvin may be isolated from the filtered broth and from the dried mycelium by organic solvent (e.g. chloroform) extraction.
