PURPOSE:To obtain a new gene useful for producing lipase by secretion of it into a culture filtrate. CONSTITUTION:A Pseudomonas bacterium-derived lipase gene having the DNA sequence of the formula. Pseudomonas cepacia IFO 14595 strain chromosome DNA is prepared and its lipase gene is cloned according to the PCR method. The primer for that treatment is selected from, e.g. a part of a known lipase in which the amino acid sequence is satisfactorily reserved. A recombinant DNA containing the lipase gene is obtained from a Pseudomonas cepacia IFO 14595 strain chromosome DNA library by using a DNA fragment containing a part of the lipase gene as a probe for screening and the sequence of the DNA is determined. |
