Mumps vaccine is prepared by attenuating a live virulent mump virus by growing it in at least ten successive passages in embryonated hens eggs and then propagating it in chick embryo tissue culture containing tissue of 9-11 day old chick embryo, at 30-38‹C. Preferably at least ten chick embryo tissue passages are made. The virulent virus may be derived from clinical material, isolated in a suitable cell culture and adapted to chick embryo tissue culture. The virus containing fluids are harvested, preferably stabilised with e.g. sucrose, human albumin, glutamine, phosphate or mixtures thereof, and then stored frozen for subsequent use as a vaccine, or filled into ampules, and freeze-dried for reconstitution with sterile water to provide a vaccine.</p> |
