摘要 PROBLEM TO BE SOLVED: To provide a novel and useful nucleic acid that can be used for detecting ethylene dichloride-decomposing bacterium and preferentially hybridizes with 16S rRNA or rDNA, and also provide a method of detecting the ethylene dichloride-decomposing bacterium and a method of decomposing ethylene dichloride and ethane chloride. SOLUTION: The objective nucleic acid comprises 18-25 nucleotide that preferentially hybridizes with 16S rRNA or rDNA of the ethylene chloride-decomposing bacterium and can be used as a primer including a base sequence bearing a specific sequence, a base sequence including the base sequence in more than 90% homology with these base sequences and the base sequence complementary to these base sequences. This invention further provides a method of detecting the ethylene chloride-decomposing bacterium in which the PCR process is carried out using the nucleic acid in the specimen as a template to detect the synthesized DNA fragment and provide a method of decomposing the ethylene chloride and ethane chloride by introducing the ethylene chloride-decomposition bacterium detected by this method into the contaminated soil and underground water.
