发明名称 ANTIBODY FOR MEASURING IMMUNOREACTION AND ELECTROCHEMICAL MEASURING METHOD USING THE SAME
摘要 PROBLEM TO BE SOLVED: To conduct the high sensitivity measurement of immunoreaction by simple and easy operation by using an antibody chemically modified with a redox active polymer and measuring the amt. of an antigen electrochemically by a finely processed minute electrode. SOLUTION: Ethylene oxide and a redox active compd. of which the redox potential is 0-1.0V to a standard hydrogen electrode such as phenothiazine are subjected to ring openingpolymerization to obtain redox active polyethylene dioxide. The mol.wt. thereof is pref. 500-2000. This compd. is reacted with an amino group of immunoglobulin, for example, by using succinic anhydride and N-hydroxysuccinimide and the reaction product is purified to obtain a bioconjugate. This bioconjugate and an antigen are mixed and reacted on a mutually penetrating comb-shaped minute electrode and the potential of the electrode is set to potential oxidizing and reducing a redox active group. The amt. of the antigen can be directly determined on the basis of current quantity. Separation and washing operations after mixing are unnecessary and high sensitive measurement is possible.
