PURPOSE:To obtain a new tyrosine phosphatase, a monoclonal antibody specifically reactive with this tyrosine phosphatase, and a hybridoma capable of producing this monoclonal antibody. CONSTITUTION:The molecule is a new tyrosine phosphatase having a molecular weight of 78kd of membrane-penetrating type and plays a role in giving lymphocytes signals. The monoclonal antibody is specifically reactive with this molecule. This monoclonal antibody is obtained by the following processes: a rodent splenocytes immunosensitized with a peptide having a sequence common with that of the tyrosine phosphatase and mouse myeloma cells are fused into hydridomas, which are then sorted to select a hydridoma capable of producing the desired antibody to make the hybridoma monocloned, which is, in turn, cultured, and from the resultant supernatant, the objective monoclonal antibody is isolated. |
