PURPOSE:To enable industrial utilization of urease for the decomposition of urea in a fermented food such as liquor, by culturing a microbial strain of Lactobacillus genus and collecting a urease having strong activity in acidic region. CONSTITUTION:Lactobacillus fermentum TK1214 (FERM P-9136) is cultured in stationary state in a medium added with meat extract, peptone, glucose, maltose, a potassium salt, sodium acetate, etc., and further with an iron salt, a manganese salt, a nickel salt, etc. The obtained cells are collected and an acidic urease is extracted from the cell. The enzyme is purified by solvent precipitation, salting-out, chromatography, etc., and is used as a crude enzyme liquid, refined enzyme liquid, dried enzyme, immobilized enzyme, etc. |
