<p>1327552 Benzylpyrimidone derivatives MORTON-NORWICH PRODUCTS Inc 21 March 1972 [25 March 1971] 13248/72 Heading C2C Novel compounds of Formula I in which R is substituted in the 3- or 4-position of the phenyl group and is hydrogen, C 1-8 alkyl, carbomethoxy, carboxy, carboxamido, dimethylamino, or aminomethyl when substituted in the 4-position or is trifluoromethyl when substituted in the 3-position, and R 1 is phenyl or hydrogen provided that R and R 1 are not simultaneously hydrogen are prepared by hydrogenation of the novel compounds of formula wherein R 2 is R or cyano which are prepared by reacting 2-hydroxypyrimidine with a compound of formula wherein X is halogen. Intermediates are prepared as follows: p-octylbenzyl chloride is obtained by action of thionyl chloride on p-octylbenzyl alcohol prepared by reduction of p-octylbenzoic acid; p-dimethylaminobenzyl chloride hydrochloride is obtained by action of thionyl chloride on the alcohol; and 1 - p - cyanobenzyl) - 2 (1H) - pyrimidone is obtained by reactingα-bromo-p-toluonitrile with 2-hydroxypyrimidine. Pharmaceutical compositions in conventional forms for per oral administration and having anti-convulsant and tranquillizing activity, comprise a novel compound of Formula I above and a carrier therefor.</p> |
