It relates to a method for obtaining and amplifying a polynucleotide whose amount present in a given sample (tester) is higher than that in another sample (driver) easily, in a short time and efficiently and a method for identifying a gene mutation in the tester using the method, which comprise the steps of (1) forming a double-stranded polynucleotide of a single-stranded polynucleotide derived from the tester and a single-stranded polynucleotide derived from the driver; and (2) removing the double-stranded polynucleotide forming a hybrid by an enzyme treatment.
