PURPOSE: To smoothly assay the presence or absence or the number of specific bacteria in a specimen not only in 'species' level but also in a 'strain' level in a simple operation in a short time. CONSTITUTION: A DNA fragment specific to a target bacterium is found by the replication, amplification and analysis using a random primer, the base sequence of the DNA fragment is determined, and based on the base sequence, a specific primer capable of selectively replicating and amplifying the DNA fragment is synthesized; using the specific primer, a DNA mixture extracted from bacteria in a specimen is replicated and amplified, and then analyzed to examine whether the specific DNA fragment is contained or not, thus assaying the presence/absence of the target bacteria in the specimen. This assaying method is combined with MPN method to assay the number of the target bacteria in the specimen. Thereby, tests can be conducted not only in closed laboratories but also outdoors, etc. |
