Humanized antibodies against TL1A

Disclosed are humanized antibodies that bind specifically to TNF superfamily member 15 (TNFSF15), also known as TL1A. Methods of making and using the anti-TL1A antibodies are also described. The humanized antibodies may be antagonists and may used to treat or diagnose conditions associated with TL1A function.

1. An isolated polynucleotide encoding an antigen-binding polypeptide that binds to specifically to TL1A, comprising:
(a) a humanized heavy chain variable region comprising:
(1) a CDR-H1 comprises the amino acid sequence selected from the group consisting of LYGMN (SEQ ID NO:11), NYGMN (SEQ ID NO:12), and TSNMGVV (SEQ ID NO:7);(2) a CDR-H2 comprises the amino acid sequence of (WINTYTGEPTYADDFKG) (SEQ ID NO:13) or HILWDDREYSNPALKS (SEQ ID NO:8); and(3) a CDR-H3 comprises the amino acid sequence selected from the group consisting of DTAMDYAMAY (SEQ ID NO:14), DYGKYGDYYAMDY (SEQ ID NO:15), and MSRNYYGSSYVMDY (SEQ ID NO:9); and/or (b) a humanized light chain variable region comprising:
(1) a CDR-L1 comprises the amino acid sequence selected from the group consisting of KSSQNIVHSDGNTYLE (SEQ ID NO:16), RSSQSIVHSNGNTYLD (SEQ ID NO:17) and SASSSVNYMH (SEQ ID NO:35);(2) a CDR-L2 comprises the amino acid sequence of KVSNRFS (SEQ ID NO:18) or STSNLAS (SEQ ID NO:36); and(3) a CDR-L3 comprises the amino acid sequence of FQGSHVPLT (SEQ ID NO:19) or HQWNNYGT (SEQ ID NO:37).
