Plates for use in thin layer chromatography, suitably made of glass or synthetic resin, carry one or more concave grooves 13, 14 adjacent one edge or two opposite edges. The plates are coated with a uniformly planar layer of absorbent 12, suitably of silica with calcium sulphate as binder, so that absorbent wholly fills the grooves which are preferably 0.5-1.5 cm. wide and 1-3 mm. in maximum depth. In use, material to be analyzed is applied along the line of a groove or along a line parallel thereto between the groove and an adjacent edge; the chromatogram may then be developed directly by standing the plate in a shallow tray of solvent with the groove parallel to the surface (see Fig. 3, not shown). Where undesired debris is present, however, a volatile solvent may first be used to establish a new origin front, after which the volatile solvent is allowed to evaporate and the plate developed as before. This latter technique is of value in estimating pesticide and herbicide residues from macerated animal and plant residues. |
