PROBLEM TO BE SOLVED: To obtain the subject new protein, having activities of a platelet- activating factor acetyl hydrolase represented by a specific amino acid sequence and useful as a preventing and a therapeutic agents for various diseases caused by the platelet-activating factor, a biochemical reagent, etc. SOLUTION: This platelet-activating factor acetyl hydrolase is a new protein having activities of a human platelet-activating factor(PAF) acetyl hydrolase represented by an amino acid sequence or an amino acid sequence having the homology therewith and is useful as a preventing and a therapeutic agents for diseases such as asthma, exudative otitis media, hemorrhagic colitis, adult respiratoty distress syndrome, etc., caused by the PAF, a biochemical reagent, etc. The protein is obtained by cloning a human fetal hepatic cDNA library and a human cerebral cDNA library with a DNA prepared by incorporating a fluorescein-labeled nucleoside using a bovine PAF acetyl hydrolase cDNA as a template according to a polymerase chain reaction(PCR), integrating the resultant cDNA into an expression vector and expressing the cDNA. |
