METHOD FOR CULTIVATION OF PROTOPLAST

The method of incubating protoplast to induce callus or cell cluster comprises: (a) putting oryza sativa cultivar, Nihonbare in 70% ethanol solution for 1 min and in sodium hypochlorite solution for 15 mins; (b) washing Nihonbare with sterilized distilled water; (c) inoculating the washed Nihonbare in agar medium containing 0.3% of casein hydrosate, 2 ppm of 2,4-D and 1 ppm of benzyladenine; (d) incubating Nihonbare at 26≦̸C for 3 weeks; (e) suspending the obtained callus in liquid medium containing 0.3% of casein hydrosate and 1 ppm of 2,4-D; (f) putting the callus in the solution containing mannitol at 27≦̸C for 6 hrs.
