PURPOSE:To obtain the subject enzyme capable of giving L-menthol for perfumes from L-menthone in markedly high productivity by immobilizing a 3-alpha- hydroxysteroid dehydrogenase on, e.g., a polymer gel carrier having tertiary (quaternary) amino group. CONSTITUTION:3-alpha-hydroxysteroid dehydrogenase is dissolved in, e.g., a phosphoric acid buffer solution (pH7.0) and a polymer gel carrier having tertiary or quaternary amino group (e.g., diethylaminoethyl cellulose) is added in the presence of NADH followed by being left to stand at room temperature for 30min and then filtration, thus obtaining the objective immobilized 3-alpha- hydroxysteroid dehydrogenase. Then, this enzyme is suspended in a phosphoric acid buffer solution (pH7.0) followed by adding a L-menthone contg. solution and shaking at 30 deg.C for 2 days and then separating the resultant product from the oil phase, thus obtaining the other objective L-menthol. |
