NEW MATERIAL:Glycoproteins A, B, C and D existing in the serum of patient suffering from hepatic cancer, mammary cancer, gastic cancer or lung cancer and having the following properties respectively: Glycoprotein A has a peak at the fraction of 1,100,000+ or -100,000 molecular weight and 4.0+ or -0.2 isoelectric point; glycoprotein B has a peak at the fraction of 900,000+ or -100,000 molecular weight and 4.0+ or -0.2 isoelectric point; glycoprotein C has a peak at the fraction of 700,000+ or -100,000 and 4.8+ or -0.2 isoelectric point; glycoprotein D has 120,000+ or -20,000 molecular weight and 6.5+ or -0.5 isoelectric point. USE:A diagnostic for the serum of cancer such as hepatic cancer, mammary cancer, gastic cancer or lung cancer. PREPARATION:For example, the glycoproteins are obtain by cultivating a cell strain NBT-2 of the cancer of the human bladder in 10% fetal bovine serum-containing culture medium, separating the cell with a centrifugal separator, bringing the culture supernatant in contact with ConA-cephalos beads to subject the glycoprotein to adsorptive separation and eluting the absorbate with a alpha-methylmannose-containing aqueous solution. These antigen are bonded to at least one of mouse monoclonal antibodies N2421, N7072 and N9261.
|
