PROBLEM TO BE SOLVED: To efficiently identify the subject fungus by amplifying DNAs in the middle region of an 18S-liposome RNA of yeast by polymerase chain reaction with a specific primer using a genome DNA of a fungus to be tested as a template, and then determining base sequences of the amplified DNAs. SOLUTION: DNAs in the middle region of a gene encoding an 18S-liposome RNA of yeast is amplified by polymerase chain reaction using a genome DNA extracted from fungus to be tested as a template, using a pair of oligonucleotides consisting of a combination of the oligonucleotide of formula I and the oligonucleotide of formula II or a combination of the oligonucleotide of formula III and the oligonucleotide of formula IV as a primer. Base sequences of the obtained amplified products are determined by a routine procedure, the obtained sequences are compared with known DNA base sequences, and then the fungus to be tested is identified as the yeast-like fungus to which the test fungus has the highest homology. The method can easily identify a yeast-like fungus for investigating the causes of diseases, the deterioration of food or the like, elucidating their propagation passes, or the like. |
