摘要 Nanolipidic Particles (NLPs) having average mean diameters of 1 nm to 20 nm are made from a precursor solution. NLPs can be loaded with a desired passenger molecule. Assemblies of these particles, called NLP assemblies, result in a vehicle population of a desired size. Single application or multifunction NLP assemblies are made from the loaded NLPs and range in size from about 30 nm to about 200 nm. A method of using preloaded NLPs to make larger carrier vehicles or a mixed population provides increased encapsulation efficiency. NLPs have application in the cosmetics, pharmaceutical, and food and beverage industries.

A method for making a nanolipidic particle (NLP) population, comprising the steps of: a. preparing a hydrous monophasic precursor solution having phospholipids having phosphatidylcholine (PC), a first concentration of an aqueous solution and a first concentration of non-aqueous solvent selected from ethanol, 1-propanol and 2-propanol; b. adding to said hydrous monophasic precursor solution one or more passenger molecules selected from the group consisting of lipophilic and amphipathic molecules to form a loaded vehicle population of passenger-loaded particles; c. diluting said loaded vehicle population with one or more dilutions of a second concentration of non-aqueous solvent at a ratio of about 1 part passenger-loaded particles to about 20 parts solvent to about 1 part passenger-loaded particles to about 0.5 parts solvent to form a passenger-loaded solvent diluted precursor solution wherein the step of diluting with said non-aqueous solvent is conducted prior to the step of adding a second concentration of said aqueous solution, and d. adding a second concentration of said aqueous solution to an aliquot of said loaded particle population to form an NLP assembly population having a mean particle diameter from about 20 to 300 nm, said NLP assembly population retaining a concentration of said non-aqueous solvent from about 0.5% to about 14%.
