A funnel (2) with a sealing ring is placed periodically over the filter strip (1) unreeled from a cassette (9). 5 ml of aq. soln. of the sample (5) are mixed with 5 ml of water (7) and 20 microlitres of fluorescent dye (6) for 5 minutes. The water is sucked out (3) and the sample is rinsed twice and dried.The funnel is removed and the sample transferred to a polished plate (8) under the microscope. Fluorescence images are read by a matrix camera (10). USE/ADVANTAGE - For counting heterotrophic bacteria in water samples, larger quantities can be processed for rapid and repeatable results.
