摘要 PROBLEM TO BE SOLVED: To obtain the subject thermally stable DNA polymerase that comprises a thermally stable DNA polymerase having the DNA synthetic activity and the 3'-5' exonuclease activity, surely amplifies DNA or RNA of high accuracy in a shortened reaction time and is useful for detection of nucleic acid and the like. SOLUTION: This novel thermally stable DNA polymerase has the DNA synthetic activity and the 3'-5' exonuclease activity, a DNA synthetic rate of at least 30 bases/second, the optimal pH of 6.5-7.5 at 70 deg.C and the optimal temperature of 75 deg.C and the molecular weight of 88-90 kDa. This DNA polymerase can synthesize DNA or RNA of high accuracy in a shortened time and is useful in the detection of nucleic acid utilizing this amplification technique. This thermally stable DNA polymerase is prepared by separating the chrcmosomal DNA from an ultrathermophilic archaebacterium KOD1 strain, subjecting the separated DNA to the PCR process using a primer synthesized based on the conserved areas of the DNA polymerase originating form Pyrococcus furiosus and incorporating the resultant gene into a vector and expressing the gene in the host cells.
