Novel indole derivatives of the formula wherein A is C 2-4 alkylene; R<SP>1</SP> is H, halogen, C 1-4 alkyl, C 1-4 alkoxy, C 1-4 alkylthio, or represents two C 1-4 alkyl groups at different positions in the indole ring (4, 5, 6 or 7); R<SP>2</SP> is H, C 1-4 alkyl, phenyl or halobenzyl; R<SP>3</SP> is H, C 1-4 alkyl or phenyl; R<SP>4</SP> is OH, CN, CONH 2 , N.(C 2-4 - alkanaryl) aminomethyl, C 1-4 alkoxy, C 2-4 alkanaryl, or C 2-5 alkoxycarbonyl; and R<SP>5</SP> is benzyl, phenyl, or mono-substituted phenyl in which the substituent is halogen, C 1-4 alkyl or halomethyl and acid addition salts thereof are prepared by reacting a compound of the formula wherein Y is halogen, alkylsulphonyloxy or arylsulphonyloxy with the corresponding 4,4- disubstituted piperidine derivative. A mixture of 4-chloro- and 6-chloro-3-(3-ptolylsulphonyloxypropyl) indole is made by tosylating with p-toluenesulphenyl chloride, the corresponding mixture of alcohols resulting from the reduction with LiAlH 4 of the methyl esters obtained by esterifying 4-chloro- and 6-chloro-3- (2-carboxyethyl) indole, resulting in docarboxylating 4-chloro- and 6-chloro-2-carboxyl-3-(2- carboxyethyl) indole, which is in turn obtained by hydrolysing the corresponding ethyl esters, made by ring closure of 5-(m-chlorophenylhydrazone) - 5 - ethoxycarbonylpentanoic acid resulting from condensing 2-ethoxycarbonylcyclopentanone with diazotized m-chloroaniline. Therapeutic compositions, having tranquillizing, anti-inflammatory and cholesterol lowering activity, and suitable for oral, parenteral or rectal administration, contain as the active ingredient a novel indole derivative of the above formula or an acid addition salt thereof. |
