The present invention refers to a method for reducing the expression levels of oncological markers in colon cancer cells, the effectiveness of this method consisting in incorporating a mixture of transfection agents and an ARNi molecule formed by a short sequence of nucleotides in colon cancer cells (HCT-15), by a transfection protocol for introducing said short molecule of ARNi to said colon cancer cells, which acts over the phosphofructokinase 1 (PFK-1) associated to the glycolytic pathway in the proliferative process of said tumorous cells through which the glucose is oxidized by the cells for obtaining energy, where ARNi-PFK-1, upon being linked to ARNm of the Phosphofructokinase 1 enzyme (PFK-1), is used to degrade the same, therefore inhibiting the synthesis of the PFK-1 protein and alternating the glycolytic pathway, this may lead to the inhibition, at RNA level, of multiple molecular targets, amongst them being the oncological markers: AMPK, APC, c-MYC and K-RAS. The invention is also related to the use and application of the RNA molecule of interference (ARNi) against Phosphofructokinase 1 (PFK-1) for silencing colon cancer cells, for reducing the expression or production levels at RNA level of the oncological markers (AMPK, APC, c-MYC and KRAS). |
