PROBLEM TO BE SOLVED: To provide methods for increasing the ratio and yield of arachidonic acid (ARA) in microbial oil, in particular, methods usable on industrial scales.SOLUTION: Provided is a method for producing a microbial oil comprising a step of culturing a microorganism by a two-stage fermentation process, where in the last stage prior to the end of fermentation, the carbon source is consumed by the microorganism at a rate higher than the rate at which carbon source is added to the culture medium; the carbon source is added at ≤0.30M carbon/kg, or at a rate that limits the microbial growth. The microorganism preferentially metabolizes fats or lipids other than ARA with such a limited carbon source to increase the proportion of ARA in the cell. Then the microbial oil is recovered from the microorganism using hexane as a solvent, where the microbial oil contains at least 50% ARA and at least 90% triglyceride.SELECTED DRAWING: None |
