PURPOSE:To prepare rapidly a large amount of a desired protein, by treating a transformed cell containing a human immunoglobulin gene with cycloheximide, so that transcription of gene to mRNA and translation development of from it to protein are promoted. CONSTITUTION:In using a transformed cell containing a human immunoglobulin gene to develop the gene, firstly, human chromosome DNA is partially digested with endonuclease to give DNA fragment. Then, operation to link it to a vector is carried out, hybrid DNA into which DNA derived from human is inserted is prepared, Escherichia coli is infected with it, and a clone containing a human immunoglobulin gene is screened. It is transduced to a myeloma cell, etc. to give a transformed cell, which is treated with cycloheximide, and a gene is developed to give a human immunoglobulin. |
