A rapid assay for quantifying the amounts of interferon in liquid samples is provided. The assay employs an affinity column, containing immobilized anti-interferon antibodies, in series with a reversed-phase column. The assay is able to measure interferon concentrations as low as 1-2 micrograms/milliliter in total assay times of less than one hour. By suitably adjusting the elution gradient used with the reversed-phase column, different interferon subtypes, e.g., monomeric and oligomeric forms of alpha 2 interferon, can be separately detected and quantified. In certain preferred embodiments, errors due to non-specific binding are minimized by treating the affinity column with an albumin-containing buffer prior to each assay.
