Native hyaluronic acid or the salt thereof is deacetylated by alkali in a water-immiscible organic solvent during from 15 to 60 mins. at a temperature from 80 to 100 degrees Celsius and is subsequently subjected to acetylation with [3H]- or [14C]-acetanhydride. A tagged hyaluronic acid is subjected to a highly effective gell permeative chromotography and the molecular weight distribution as well as radioactivity are measured in thereby obtained fractions by means of the liquid scintillation spectography.
