PURPOSE:To achieve a longer life and a shorter conditioning of a column by separating optical isomers in a sample by a high-speed liquid chromatography employing a compound having a camphor sulfonate group of an optical isomerism as ion pair reagent. CONSTITUTION:A carrier liquid having an asymmetric carbon-containing compound with d-10-camphor sulfonate or the group thereof added thereto is held into a carrier liquid tank 3 while orthophthalaldehyde is held into a reagent tank 7 and the equipment is operated to condition a high-speed chromatography column 1 in a thermostatic cell 9 for 20-30min. After the conditioning, a sample is put thereinto at an injection port 8 while a carrier liquid is fed from the tank 3 and orthophthalaldehyde from the sample tank 7. The mixture is sent to a fluorescence detector 5 through a mixing coil 4 to separate and detect optical isomers labelled by orthophthalaldehyde individually. This enables a longer life as well as shorter conditioning of the column without taking harmful components into a column filler. |
