PURPOSE:To enable high-yield production of bacterial cellulose which is useful in the fields of foods, cosmetics and coatings by culturing a novel pyrroloquinolinequinone(PQQ)-unproductive strain belonging to cellulose- generating Acetobacter and recovering the product from the culture products. CONSTITUTION:Acetobacter sp. BPR 2001 strain (FERM-BP-4545) is inoculated into a medium and subjected to static culture at 28 deg.C for 3 days. Then, the culture mixture is centrifuged to collect the cells, which are suspended in a phosphate buffer containing nitrosoguanidine(NTG) to effect the chemical mutation treatment and screen the PQQ-unproductive strain whereby a novel PQQ- unproductive strain BPR 3001C is isolated. This strain is inoculated into the medium and cultured at 28 deg.C for 4 days to accumulate cellulosic substance in the medium, then the substance is collected whereby the cellulosic substance is produced economically in high yield. |
