PURPOSE:To obtain a new monoclonal antibody recognizing the C-terminal part of vitamin D receptor, not having the vitamin D bound thereto as well as existing on a nuclear membrane and in a cytoplasm, and useful for elucidating the action mechanism of the vitamin D. CONSTITUTION:This monoclonal antibody recognizing an amino acid sequence near to the C-terminal of the human vitamin D receptor(VDR), the VDR not having the vitamin D bound thereto, and the VDR existing on a nuclear membrane and in a cytoplasm, is obtained by synthesizing a peptide corresponding to the No.338-352 amino acid sequence of the VDR by a conventional peptide synthetic method, binding the synthesized peptide to a bovine serum albumin, administering the prepared immunogen and an adjuvant into a Bal b/c mouse for immunizing the mouse, collecting the splenic cells of the mouse after the final immunization of the mouse, fusing the splenic cells with mouse myeloma cells, culturing the fused cells in a HAT medium, screening the obtained hybridoma, cloning the screened cells by a limiting dilution analysis method, and finally culturing the cloned cell. |
