摘要 PURPOSE:To accurately and readily detect a nucleic acid sample useful for diagnosing viral infection, etc., by binding plural regions of a sample DNA to plural kinds of fluorescence-labeled oligonucleotides, elongating the extended chain according to enzymic reaction and then repeatedly preforming the operation. CONSTITUTION:Oligonucleotides are respectively bound to plural regions of the same or different sample DNAs to be a template and extended chains are elongated by enzymic reaction. The extended chains corresponding to the respective regions are reused as the template to repeat the reaction. Thereby, the plural regions of the sample DNAs are amplified to prepare nucleic acid samples. In the method, the oligonucleotides are at least two kinds of oligonucleotides having mutually unidentical sequences. The at least two oligonucleotides are labeled with respective different fluorescent coloring matters and the extended chains are labeled with different fluorescent coloring matters according to enzymic reaction. Thereby, nucleic acid fragments corresponding to plural regions labeled with the fluorescent coloring matters are prepared and separated according to the molecular weight. The fluorescent coloring matters are then excited with light to detect the fluorescence to detect the nucleic acid samples.
