PROBLEM TO BE SOLVED: To provide a method for analyzing nucleic acid base sequence for detecting the base mutation of DNA (SNPs), a reagent kit and a nucleic acid base sequence-analyzing device. SOLUTION: This method for analyzing the nucleic acid base sequence is provided by pre-treating each of the reagents used for a complemental chain- elongating reaction and chemo-luminescent reaction with a pyrophosphatase and an apyrase for decomposing an impurity PPi inhibiting the chemo- luminescent reaction with the pyrophosphatase and also an impurity ATP with the apyrase, and performing the complemental chain-elongating reaction and the chemo-luminescent reaction by using the above pre-treated reagents.
