Factor H binding proteins (FHBP) with altered properties and methods of use thereof

Factor H binding proteins that can elicit antibodies that are bactericidal for at least one strain of N. meningitidis, and methods of use of such proteins, are provided.

A non-naturally occurring factor H binding protein (fHbp) comprising a non-naturally occurring amino acid sequence having at least 99% amino acid sequence identity to the amino acid sequence set forth in SEQ ID NO:2, wherein the non-naturally occurring fHbp comprises an amino acid substitution for aspartic acid at position 211 (D211) with alanine, wherein numbering of D211 is based on the numbering of amino acid residues in SEQ ID NO: 1, wherein said fHbp binds human factor H (fH) with an affinity that is 55% or less of the affinity of Neisseria meningitidis fHbp ID22 for human fH, wherein the fHbp ID22 has the amino acid sequence of SEQ ID NO:2.
