PROBLEM TO BE SOLVED: To obtain a new water-soluble hexacyclic compound having an excellent antitumor activity and useful as an antitumor medicine. SOLUTION: A compound of formula I [R1 and R2 are each H, hydroxyl, a 1-6C alkyl or the like; R3 is a 1-6C alkyl; R4 is H, (protected) amino or the like; Z is O, S, CR5R6 [R5 and R6 are each H or a 1-6C alkyl; (m) and (n) are each 0-2], for example, (9S)-1-amino-9-ethy1-5-fluoro-2,3-dihydro-3,9-dihydroxy-4- methyl-1H,12H-benzo[de]pyrano[3',4':6,7] indolizino[1,2-b]quinoline-10,13(9H,15H)- dione. The compound of formula I is obtained by administering a compound of formula II into an animal and subsequently isolating from its urine, or treating a microsome (for example, baculovirus expression system) containing a human liver metabolism enzyme with the compound of formula II and subsequently isolating from the reaction solution.
